From 1976 to 1980, many scholars studied the fermentation technology of Aureobasidium pullulans and the properties of pullulan polysaccharides, which was the first research peak. From 1984 to 1996, a large number of reports on the production mechanism and application of pullulan polysaccharides have been published, which is the second research peak. Fermentation research focuses on optimizing culture conditions, maintaining high cell productivity, shortening fermentation time, reducing production costs, and improving the purity of final products to meet the needs of food, cosmetics and medicine.

Pullulan polysaccharide is a linear extracellular polysaccharide produced by Aureobasidium pullulans. It is mainly composed of repetitive unit maltose linked by a-(1-6) glycoside bond. Its unique chemical structure gives it excellent properties, such as high water solubility, film forming, oxygen resistance, plasticity, low viscosity and biodegradable physical and chemical properties. Therefore, pullulan polysaccharides are widely used in food and medicine industries. The synthesis of pullulan by Aureobasidium pullulans is a complex physiological metabolic process, which is greatly influenced by the external environment.

Dry Pullulan polysaccharides are white and tasteless powder. When the concentration of 5-10% Pullulan polysaccharides dissolves in water, it will form a viscous solution without moisture absorption. The solution is linear, without branched chains, and curls irregularly. When the temperature reached 250 C, the pullulan polysaccharide began to decompose and carbonized at 280 C. Pullulan polysaccharides are highly soluble in water and dilute alkali solutions, except dimethyl sulfoxide and dimethylformamide, insoluble in ethanol and other organic reagents. Pullulan polysaccharide is non-toxic, mutagenic, non-carcinogenic, odorless, tasteless and edible. Pullulan polysaccharide has excellent film-forming and adhesion. The membrane has considerable mechanical strength. It can be biodegraded and has good oxygen resistance, but it can not be degraded by enzymes in human digestive tract. Pullulan aqueous solution has low viscosity, similar to Arabic gum. The viscosity of Pullulan polysaccharide solution does not change with temperature and pH, nor is it affected by most metal ions. Molecular weight of pullulan polysaccharide is related to medium conditions and strains. High molecular weight pullulan polysaccharide is more suitable for commercial use. The average molecular weight of pullulan polysaccharide is about 100-200 kDa, and the average molecular weight is about 362-480 kDa. The molecular weight dispersion index is generally between 2.1 and 4.1. These data are important reference standards for the application of pullulan polysaccharides in pharmaceutical industry.
