Introduction for Fucoxanthin analysis

China's wakame is widely cultivated, and the content of fucoxanthin in wakame is high. Therefore, in this experiment, Fucoxanthin analysis is extracted from dried wakame. At present, the extraction of Fucoxanthin analysis is mostly carried out by organic solvent extraction, ultrasonic assisted extraction and enzyme assisted extraction. The infiltration method and the reflux extraction method were used to compare the extraction rate of fucoxanthin, and the best extraction process was selected, and the difference between the two extraction methods was compared.


Comparison of extraction processes

Through visual tests, it was found that the percolation rate > solvent dosage > swelling time, in which the percolation rate and solvent dosage had a significant effect on the percolation process. The optimal extraction combination was a percolation rate of 2 mL·min -1 , a solvent usage of 12 times and a swelling time of 30 min. Statistical analysis showed that there was no significant difference in the extraction rate compared with the reflux method, but there was a significant difference in the content of Fucoxanthin analysis in the extract. The content of Fucoxanthin analysis in the extract obtained by the percolation method was higher.

Fucoxanthin analysis is unstable in nature and has a unique accumulation of diene, 5,6-epoxy structure and optical rotation, making it susceptible to isomerization and degradation by light and heat. When the reflux method is used for extraction, heating can increase the molecular motion of the solvent in the solvent, which is beneficial to the dissolution of the active material, and at the same time, the temperature can also cause degradation and isomerization of the fucoxanthin. The percolation method is carried out at room temperature, to a certain extent. Isomerization of Fucoxanthin analysis is avoided. In addition, the content of fucoxanthin in the extract obtained by the percolation method is higher than that of the reflux method, and the fucoxanthin is extracted by the percolation method.
